目的观察芪丹颗粒对博莱霉素A5诱导的肺纤维化大鼠病变严重程度及FasL、Caspase-3表达的影响,以探讨其可能的作用机制。方法70只健康雄性清洁级Sprague-Daw ley(SD)大鼠,随机分成4组。28、42d正常对照组(对照组)各5只,模型组各10只,芪丹组各10只,氢可组各10只。对照组气管内和灌胃均用生理盐水,余SD大鼠气管内灌注博莱霉素A5诱导肺纤维化,第15d开始每日分别给予芪丹颗粒剂灌胃(芪丹组3125mg/kg)、腹腔注射氢可(氢可组25mg/kg)。分别在第28、42d处死。苏木素-伊红(HE)染色观察肺泡炎及纤维化程度,免疫组化观察石蜡切片中FasL及Caspase-3的表达。结果芪丹颗粒剂组较氢可组及模型组肺纤维化及肺泡炎程度轻。FasL表达芪丹组28、42d与对照组差异有统计学意义,P<0.01;与模型组及氢可组比较差异无显著性。Caspase-3芪丹组与模型组及氢可组比较表达差异无显著性意义。结论芪丹颗粒剂可减轻博莱霉素A5诱导的肺纤维化大鼠的肺纤维化及肺泡炎。肺纤维化形成后期应用芪丹颗粒剂对FasL及Caspase-3的表达无减轻。
Objective To observe the effect of Qidan Granules on the severity of bleomycin A5-induced pulmonary fibrosis in rats and the expression of FasL and Caspase-3 in order to explore its possible mechanism. Methods Seventy healthy male Sprague-Dawley rats were randomly divided into four groups. 28, 42d normal control group (control group) 5 each, model group 10 each, Qi Dan group 10 each, Hydrochlorothiazide 10 each. Intratracheal and intragastric administration in the control group were treated with saline, while intratracheal intratracheal instillation of Bleomycin A5 induced pulmonary fibrosis in the other SD rats. From the 15th day onward, Qi Dan granules were given to the rats respectively (3125mg / kg for QiDan group) , Intraperitoneal injection of hydrogen (hydrogen group 25mg / kg). Were killed on the 28th and 42nd day respectively. The hematoxylin-eosin (HE) staining was used to observe the degree of alveolitis and the degree of fibrosis. The expression of FasL and Caspase-3 in paraffin sections was observed by immunohistochemistry. Results Qidan granules group than hydrogen group and model group, pulmonary fibrosis and alveolitis degree of light. The expression of FasL in Qidan group for 28,42 days was significantly different from that in control group (P <0.01). There was no significant difference between the model group and the hydrogen group. There was no significant difference in the expression of Caspase-3 between Qidan group and model group and hydrogen group. Conclusion Qidan granules can reduce pulmonary fibrosis and alveolitis in bleomycin A5-induced pulmonary fibrosis rats. The late application of Qidan granules in the formation of pulmonary fibrosis did not reduce the expression of FasL and Caspase-3.
