目的探索胰腺癌EGFR和HER3的基因状态、蛋白表达与胰腺癌临床病理因素之间的关系,分析它们之间的相关性,探讨EGFR和HER3在胰腺癌中的作用。方法分别采用荧光原位杂交(FISH)和免疫组织化学(IHC)方法,检测55例中国人胰腺癌及癌旁组织EGFR和HER3基因状态和蛋白表达,结合临床病理资料进行统计学分析。结果 55例胰腺癌中,EGFR(FISH)阳性33例(60.0%)。EGFR(IHC)阳性34例(61.8%)。HER3(FISH)阳性26例(47.3%)。HER3(IHC)阳性4例(7.3%)。统计结果显示,EGFR、HER3蛋白表达与基因扩增间无明显相关性(P>0.05)。EGFR与HER3之间在蛋白的表达以及基因状态上也无明显相关性(P>0.05)。EGFR、HER3蛋白表达及基因扩增与患者的性别、年龄、肿瘤大小、肿瘤分化程度、胰周浸润,肝转移均无关(P>0.05)。结论在胰腺癌中,EGFR的蛋白表达显著高于癌旁组织;HER3蛋白表达与癌旁组织无显著差异。胰腺癌中存在较高比例的EGFR、HER3基因扩增。EGFR、HER3蛋白表达与基因拷贝数增加无关。EGFR与HER3相互之间在蛋白表达及基因状态上也不相关。EGFR与HER3在胰腺癌中的作用尚不明确。
Objective To explore the relationship between the gene status and protein expression of EGFR and HER3 in pancreatic cancer and the clinicopathological factors of pancreatic cancer, and to explore the correlation between EGFR and HER3 in pancreatic cancer. Methods The fluorescence and in situ hybridization (FISH) and immunohistochemistry (IHC) methods were used to detect the expression of EGFR and HER3 in 55 cases of Chinese pancreatic cancer and paracancerous tissues, respectively. The clinical and pathological data were used for statistical analysis. Results In 55 cases of pancreatic cancer, EGFR (FISH) positive in 33 cases (60.0%). 34 cases of EGFR (IHC) positive (61.8%). HER3 (FISH) positive in 26 cases (47.3%). HER3 (IHC) positive in 4 cases (7.3%). The statistical results showed that there was no significant correlation between EGFR and HER3 protein expression and gene amplification (P> 0.05). There was no significant correlation between EGFR and HER3 in protein expression and gene status (P> 0.05). The expression of EGFR and HER3 protein and gene amplification had no correlation with the gender, age, tumor size, tumor differentiation, peripancreatic invasion and liver metastasis (P> 0.05). Conclusion The protein expression of EGFR in pancreatic cancer is significantly higher than that in paracancerous tissues. The expression of HER3 protein is not significantly different from that in paracancerous tissues. There is a high proportion of EGFR in pancreatic cancer, HER3 gene amplification. EGFR, HER3 protein expression and gene copy number has nothing to do. EGFR and HER3 are not related to each other in terms of protein expression and gene status. The role of EGFR and HER3 in pancreatic cancer is unclear.
