目的建立酸水解在中药HPLC中的应用新方法。方法 1%盐酸,99℃,3h,固液比1g:100m L,水解中药粉末。用10%氢氧化钠-甲醇溶液中和水解产物,并过滤中和液,取滤液进HPLC。考察三种中药标准品、水解前样品、水解后样品的色谱条件是否达到要求,观察峰形及峰量的变化。做了三种中药的酸解后样品含量测量的方法学研究。结果通滞苏润江胶囊、益气糖康胶囊、黄连中药材的盐酸水解的HPLC图谱质量较标准方法有很大的提高,峰形得到了优化。三种中药的酸解后样品的含量测定方法是药典符合要求的。结论 1%盐酸水解中药粉能提高一些中药的HPLC图谱质量,优化峰形。酸水解对峰形不理想的图谱质量有很大的改进,并提高中药控制质量。
OBJECTIVE To establish a new method for acid hydrolysis in HPLC of traditional Chinese medicine. Method 1% hydrochloric acid, 99 ℃, 3h, solid-liquid ratio 1g: 100m L, hydrolyzed Chinese medicine powder. The hydrolyzate was neutralized with 10% sodium hydroxide-methanol solution and the neutralized solution was filtered and the filtrate was taken to HPLC. To investigate whether the chromatographic conditions of the three traditional Chinese medicine standards, prehydrolysis samples and hydrolyzed samples meet the requirements, and observe the changes of peak shape and peak amount. A methodological study was conducted to measure the content of three kinds of traditional Chinese medicines after acid hydrolysis. Results The results showed that the quality of HPLC chromatogram of Tongli SuRunJiang Capsule, Yiqitankang capsule and Coptis chinensis was significantly improved compared with the standard method, and the peak shape was optimized. The determination of the content of the three traditional Chinese medicines after acid hydrolysis is in accordance with the requirements of the pharmacopoeia. Conclusion 1% hydrochloric acid hydrolyzed Chinese medicine powder can improve the HPLC chromatogram of some traditional Chinese medicines and optimize the peak shape. Acid hydrolysis of the peak shape is not ideal for the quality of the map has greatly improved, and improve the quality of Chinese medicine control.
