将平消胶囊灌喂大鼠,制备含药血清,血清培养胆管癌细胞,采用微管吸吮技术,研究不同剂量的含药血清对胆管癌细胞黏弹性以及黏附力学特性的影响。结果显示,胆管癌细胞具有高弹性系数和低黏性系数的黏弹性特征,高剂量和中剂量含药血清处理后的胆管癌细胞黏弹性参数K1、K2和μ值较对照组均明显降低(P<0.01),低剂量变化不明显;细胞与CD44v6单克隆抗体的黏附力随着药物剂量的增加而显著降低(P<0.01)。以上结果提示平消胶囊破坏了胆管癌细胞骨架,抑制细胞膜上黏附分子CD44v6表达,降低癌细胞与血管内皮细胞的黏附几率,阻止胆管癌细胞转移。
The rats were administrated with Pingxiao capsule to prepare the drug-containing serum and cholangiocarcinoma in serum. Microtubule sucking technique was used to study the effect of different doses of drug-containing serum on the viscoelasticity and adhesiveness of cholangiocarcinoma cells. The results showed that the cholangiocarcinoma cells had the characteristics of high elasticity and low viscoelasticity. The viscoelasticity parameters K1, K2 and μ of cholangiocarcinoma cells treated with high dose and medium dose of serum were significantly lower than those of the control group P <0.01). The adhesion of CD44v6 cells to CD44v6 monoclonal antibody was significantly decreased with the increase of drug dose (P <0.01). The above results suggest that Pingxiao capsule destroys the skeleton of cholangiocarcinoma, inhibits the expression of adhesion molecule CD44v6 on the cell membrane, reduces the adhesion of cancer cells and vascular endothelial cells, and prevents the metastasis of cholangiocarcinoma cells.
