目的利用大鼠被动型Heymann肾炎(PHN)模型,研究冬虫夏草提取物——多球壳素(ISP-1)对膜性肾病的疗效。方法雄性SD大鼠24只随机分为正常对照组、PHN模型组及ISP-1干预组,每组各8只。PHN模型组及ISP-1干预组均经腹腔单次注射抗大鼠近端肾小管刷状缘抗体(FXIA)血清(5 mL·kg-1)制作模型。ISP-1干预组在制作模型同时,予ISP-1 25 mg·kg-1.d-1干预。定期检测其24 h尿蛋白定量及血液生化指标(血清总蛋白、清蛋白、总胆固醇及肌酐),并于第4周末取其肾皮质标本切片行光镜、免疫荧光及电镜检查。结果正常对照组大鼠24 h尿蛋白定量、血液生化指标及肾脏病理均无异常。PHN模型组及ISP-1干预组大鼠均出现大量蛋白尿及血胆固醇增高,免疫荧光检查发现2组肾小球内均有沿毛细血管壁的免疫复合物沉积,电镜发现2组肾小球上皮下均有电子致密物沉积。与PHN模型组比较,ISP-1干预组尿蛋白定量及血胆固醇均明显减轻(Pa<0.01),病理改变亦有改善。结论 ISP-1能明显减少尿蛋白,调节脂代谢,且能有效缓解PHN肾脏损伤,是冬虫夏草治疗膜性肾病发挥功效的重要物质基础。
Objective To investigate the curative effect of Cordyceps sinensis extract - polysaccharide (ISP-1) on membranous nephropathy by using rat passive Heymann nephritis (PHN) model. Methods Twenty-four male SD rats were randomly divided into normal control group, PHN model group and ISP-1 intervention group, with 8 rats in each group. PHN model group and ISP-1 intervention group were injected intraperitoneally with anti-rat proximal tubular brush border antibody (FXIA) serum (5 mL · kg-1) model. ISP-1 intervention group at the same time as the model, to ISP-1 25 mg · kg-1.d-1 intervention. The 24 h proteinuria and blood biochemical parameters (serum total protein, albumin, total cholesterol and creatinine) were measured regularly. The specimens of renal cortex were taken by light microscope, immunofluorescence and electron microscopy at the end of the 4th week. Results The normal control group 24 h urinary protein, blood biochemical parameters and kidney pathology no abnormalities. PHN model group and ISP-1 intervention group rats showed a large amount of proteinuria and elevated blood cholesterol, immunofluorescence showed that the glomerular capillary within the immune complex deposition in both groups, electron microscopy found that two groups of glomeruli Under the epithelium are electron dense deposits. Compared with PHN model group, proteinuria and blood cholesterol of ISP-1 intervention group were significantly reduced (Pa <0.01), and pathological changes were also improved. Conclusion ISP-1 can significantly reduce urinary protein, regulate lipid metabolism, and can effectively alleviate PHN kidney injury, is Cordyceps sinensis treatment of membranous nephropathy play an important material basis.
