目的 观察壬基酚对大鼠睾丸抑制素和雄激素结合蛋白的影响。方法 将 36只 19d龄SD雄性大鼠分为 4组 ,分别腹腔 1次注射 0、11、4 4和 176mg/kg壬基酚后 ,于 6h、2 4h和 4 8h每组各处死 3只动物 ,取睾丸提取RNA后 ,用RT -PCR法对抑制素和雄激素结合蛋白的表达量进行定量分析。结果 雄激素结合蛋白基因的表达量随所给NP剂量的增高而降低 ,呈剂量效应关系。与对照组比较 ,在 6h各组中 ,ABP的表达量降低均具有高度显著性 (P <0 .0 1) ;在 2 4h各组中 ,与对照组比较 ,4 4和 176mg/kg组的ABP表达降低有显著性 (P <0 .0 5或 <0 .0 1)。在 4 8h各组中 ,仅 176mg/kg组的降低具有显著性 (P <0 .0 5 )。 2 4和 4 8h抑制素表达量 ,在 4 4和 176mg/kg剂量组降低 ,与对照组比较有统计学意义 (P <0 .0 1)。结论 壬基酚可以抑制大鼠睾丸Sertoli细胞雄激素结合蛋白的表达。低剂量的壬基酚可能并不影响Sertoli细胞抑制素的表达 ,但高剂量时 ,可以使抑制素表达降低
Objective To observe the effects of nonylphenol on testis and androgen binding protein in rats. Methods Thirty-six male Sprague-Dawley rats, aged 19 days, were divided into 4 groups. One animal was injected intraperitoneally with 0, 11, 44 and 176 mg / kg of nonylphenol. Three animals were sacrificed at 6h, 24h and 48h respectively The testis was used to extract RNA, and the expression of inhibin and androgen-binding protein was quantitatively analyzed by RT-PCR. Results The expression of androgen binding protein gene decreased with the increase of NP dose, showing a dose-response relationship. Compared with the control group, in the 6h groups, the expression of ABP was significantly decreased (P <0.01); in the 24 h groups, compared with the control group, 44 and 176 mg / kg groups ABP expression decreased significantly (P <0.05 or <0.01). In the 48 h groups, only the 176 mg / kg reduction was significant (P <0.05). The expression of inhibin at 24 h and 48 h decreased in the 4 4 and 176 mg / kg groups, which was statistically significant compared with the control group (P <0.01). Conclusion Nonylphenol can inhibit the expression of androgen binding protein in Sertoli cells of rats. Low doses of nonylphenol may not affect Sertoli cytostat expression, but at high doses, inhibin expression may be reduced
