为探讨生长抑素(SST)对内皮素(ET)刺激细胞增殖的作用及其机理,本工作在培养的兔主动脉血管平滑肌细胞(VSMC)上发现,10 ̄(-8)mol/LET刺激细胞增殖(3N-TdR参入增多)和丝裂素活化蛋白激酶(MAPK)激活,10 ̄(-8)mol/LSST单独作用可抑制细胞增殖但不影响MAPK活性。SST不仅呈浓度依赖性地抑制ET刺激的VSMC增殖(P<0.01),而且亦抑制ET刺激的细胞MAPK激活(P<0.01)。结果表明,SST具有抑制ET刺激的MAPK系统激活的作用,且这种作用可能是发生在MAPK的上游环节,这可能是其抑制ET促细胞增殖的机理之一。
In order to investigate the effect and mechanism of somatostatin (SST) on endothelin (ET) stimulated cell proliferation, we found that stimulation of 10 -8 mol / L ET in cultured rabbit aortic vascular smooth muscle cells (VSMCs) Cell proliferation (3N-TdR incorporation increased) and mitogen-activated protein kinase (MAPK) activation, 10 ~ (-8) mol / L.LSST alone can inhibit cell proliferation but does not affect MAPK activity. The results showed that SST could inhibit the activation of MAPK stimulated by ET, and this effect may occur in the upstream of MAPK, which may be one of the mechanisms of inhibiting the proliferation of ET-stimulated cells.
