目的:建立安神补心丸(丹参、五味子、石昌蒲等)中五味子醇甲、五味子酯甲和五味子甲素含量的HPLC测定方法。方法:样品用超声提取,过滤,取续滤液,采用RP-HPLC法直接测定,用十八烷基键合相,甲醇-水-冰醋酸(65∶35∶0.1)为流动相,流速为1 mL/min,检测波长为220 nm。结果:五味子醇甲、五味子酯甲,五味子甲素分别在0.001 2~0.500 0μg(r=1.000 0)、0.001 4~0.576 0μg(r=0.999 5)和0.002 1~0.823 0μg(r=0.999 8)范围内,质量与峰面积具有良好线性关系。平均加样回收率分别为99.51%,99.40%和99.06%,平均RSD(n=5)分别为1.48%、1.27%和1.38%。方法用于安神补心丸中五味子醇甲、五味子酯甲和五味子甲素的含量测定,RSD(n=5)分别为1.19%~1.53%、1.09%~1.52%和1.36%~1.72%。结论:方法具有灵敏度高,准确,简便,稳定快速的特点,能用于安神补心丸中五味子醇甲、五味子酯甲、五味子甲素的含量测定。
Objective: To establish an HPLC method for determination of schisandrin, schisandrin and schizandrin in Anshenbuxin Pills (Salvia miltiorrhiza, Schisandrin, Shichangpu, etc.). Methods: The sample was extracted by ultrasound, filtered, and the filtrate was taken. The filtrate was directly determined by RP-HPLC. The octadecyl bonding phase was used. The mobile phase consisted of methanol-water-glacial acetic acid (65:35:0.1) and the flow rate was 1. mL/min, detection wavelength is 220 nm. Results: Schisandra arborifolium, schisandrin A, and schisandra A were 0.001 2 to 0.500 0 μg (r=1.000 0), 0.001 4 to 0.576 0 μg (r=0.999 5) and 0.0021 to 0.823 0 μg (r=0.999 8), respectively. Within the range, there is a good linear relationship between mass and peak area. The average sample recovery was 99.51%, 99.40% and 99.06%, respectively, and the average RSD (n=5) was 1.48%, 1.27%, and 1.38%, respectively. The method was used to determine the contents of Schisandrin, Schizandrin A and Schisandrin A in Anshen Buxin Pills. RSD (n=5) was 1.19%-1.53%, 1.09%-1.52%, and 1.36%-1.72%, respectively. Conclusion: The method has the characteristics of high sensitivity, accuracy, convenience, stability and rapidity. It can be used to determine the content of Schisandrin, Schizandrin, and Schizandrin in Anshen Buxin Pills.
