本研究应用Dot-ELISA方法,以卫氏并殖吸虫(Pw)成虫单克隆抗体A_3D_3(McAbA_3D_3)检测5只实验感染犬吡喹酮治疗前后血清循环抗原(CAg)动态。结果表明,检出CAg最早时间为感染后第4d,至第19d,4只犬CAg检测均为阳性。感染后第101d,其中2只以吡喹酮总量3.2g进行治疗,治疗后短期内CAg滴度明显升高,至治疗后第6d达高峰,1号犬和3号犬分别在治疗后第65d和26dCAg滴度下降为0。2只未治疗感染犬CAg滴度维持于1:160~1:640。治疗后80d剖检2只治疗犬,其肺部均有多个陈旧性虫囊,内多为变性坏死虫卵,未发现虫体。未治疗犬于死后解剖获成虫60条。上述研究结果提示用McAbA_3D_3作Dot-ELISA对犬的Pw活动性感染具有早期诊断和疗效考核价值。
In this study, Dot-ELISA method was used to detect the activity of CAg in 5 experimental infected dogs before and after treatment with pAQ3 monoclonal antibody A_3D_3 (McAbA_3D_3). The results showed that the detection of CAg earliest time after infection 4d, to 19d, 4 dogs CAg test were positive. At 101 days after infection, two of them were treated with praziquantel in a total amount of 3.2 g. The titer of CAg increased significantly in the short term after treatment, reaching the peak on the 6th day after treatment. The first and the third dogs were treated with The titers of 65 and 26 dCAg dropped to 0.2 and the CAg titer of untreated dogs was maintained at 1: 160-1: 640. After 80 days of treatment, two dogs were dissected, and their lungs had multiple old bladders. Most of them were degenerative and necrotic eggs, and no parasites were found. Untreated dogs were dissected 60 adult worms. The above results suggest that the use of McAbA_3D_3 for Dot-ELISA in dogs with Pw active infection has early diagnosis and efficacy evaluation.
