目的探讨贝伐珠单抗对肝癌细胞HepG2血管内皮生长因子(vascular endothelial growth factor,VEGF)相关受体的影响。方法通过不同浓度的贝伐珠单抗降低培养液中VEGF来模拟低浓度VEGF的肿瘤微环境,RT-PCR、ELISA、Western blotting检测肿瘤细胞VEGF相关受体表达水平的变化。结果细胞培养液的VEGF浓度大幅度下降(P<0.05);可溶性VEGFR2的浓度出现了上升(P<0.05);HepG2细胞VEGF和VEGFR1表达水平无明显变化(P>0.05);VEGFR2表达水平出现了明显的上调(P<0.05)。结论贝伐珠单抗导致了肝癌细胞HepG2大幅度上调了VEGFR2的表达。
Objective To investigate the effect of bevacizumab on hepatocellular carcinoma HepG2 vascular endothelial growth factor (VEGF)-related receptors. Methods The tumor microenvironment of low concentration of VEGF was simulated by reducing VEGF in the culture medium with different concentrations of bevacizumab. The expression of VEGF-related receptors in tumor cells was detected by RT-PCR, ELISA and Western blotting. Results The VEGF concentration in cell culture medium decreased significantly (P<0.05); the concentration of soluble VEGFR2 increased (P<0.05); the expression levels of VEGF and VEGFR1 in HepG2 cells did not change significantly (P>0.05); the expression level of VEGFR2 appeared. Significantly upregulated (P < 0.05). Conclusion Bevacizumab induced hepatocellular carcinoma cell HepG2 to significantly upregulate the expression of VEGFR2.
