目的应用MTT比色法比较六种抗肿瘤药物对脑胶质瘤细胞的抑制作用,为临床化疗筛选药物、制定化疗方案提供指导。方法应用MTT比色法对62例脑胶质瘤新鲜标本进行化疗药物敏感性研究。结果六种药物敏感率经卡方检验有明显差别,由高至低依次为:Vm-26、Fotemustine、BCNU、CBP、DDP、Vp-16。病人性别对药物敏感率无影响,年龄、术前机能状态KPS评分对大多数药物敏感率无影响,肿瘤病理分级只对部分药物敏感率有影响。结论MTT比色法是一种快速、准确检测体外药物敏感性的方法。Vm-26抑制肿瘤细胞增殖作用较强,优于Fotemustine、BCNU、CBP、DDP、Vp-16。
Objective To compare the inhibitory effects of six anti-tumor drugs on glioma cells by MTT assay and to provide guidance for the screening of drugs and the development of chemotherapy regimens. Methods MTT colorimetric method of 62 cases of glioma fresh specimens for chemosensitivity studies. Results The sensitivities of the six drugs were significantly different by chi square test. The highest to the lowest were Vm-26, Fotemustine, BCNU, CBP, DDP and Vp-16. The gender of the patient has no effect on the sensitivity of the drug. KPS scores of age and preoperative functional status have no effect on the sensitivity of most drugs. The pathological grade of the tumor only affects the sensitivity of some drugs. Conclusion MTT colorimetric method is a rapid and accurate method to detect in vitro drug sensitivity. Vm-26 inhibited tumor cell proliferation stronger than Fotemustine, BCNU, CBP, DDP, Vp-16.
