目的建立高效液相色谱法同时测定山蜡梅中东莨菪素、槲皮素、夏蜡梅碱量的方法。方法HypersilODS柱(250mm×4.6mm,25μm);流动相为甲醇-水,梯度洗脱;检测波长为348、376、222nm;体积流量为1mL/min。结果山蜡梅叶中东莨菪素、槲皮素、夏蜡梅碱达到基线分离且线性关系良好,线性范围为10.02~50.10、10.04~50.20、10.04~50.20μg/mL,平均加样回收率分别为97.13%、96.68%、97.22%。结论本方法用于山蜡梅中东莨菪素、槲皮素、夏蜡梅碱的定量测定,具有快捷、简便、准确的特点,能够多成分综合评价该药材。
OBJECTIVE To establish a method for the simultaneous determination of midodrophyllin, quercetin and simethicone by HPLC with high performance liquid chromatography. Methods The Hypersil ODS column (250 mm × 4.6 mm, 25 μm) was used. The mobile phase was methanol-water with gradient elution. The detection wavelength was 348,376,222 nm. The volume flow rate was 1 mL / min. Results The results showed that there was a good linear relationship between the isolates of Scutellariae Setaria, quercetin and sarcocalyxes from the baseline in the linear range of 10.02 ~ 50.10,10.04 ~ 50.20,10.04 ~ 50.20μg / mL. The average recoveries were 97.13%, 96.68%, 97.22%. Conclusion The method is suitable for the quantitative determination of scopolamine, quercetin and sarcocalyxin in the Chinese yam. It has the characteristics of quickness, simplicity and accuracy, and can be used for the comprehensive evaluation of the medicinal materials.
