目的分析鉴定皱纹盘鲍脏器糖蛋白AGP-Ⅲ初级结构并探讨脏器糖蛋白粗品AGP的免疫活性。方法采用高效液相色谱法、氨基酸组成分析法、红外光谱法、β-消除反应对AGP-Ⅲ进行初步分析;建立小鼠免疫低下模型,探讨AGP不同剂量对小鼠免疫功能的调节作用。结果AGP-Ⅲ的单糖组成有岩藻糖、半乳糖、氨基葡萄、甘露糖、氨基半乳糖、木糖、葡萄糖和鼠李糖。AGP-Ⅲ含有17种氨基酸,缬氨酸含量最高,其次为谷氨酸、甘氨酸。AGP-Ⅲ中存在糖蛋白的特征吸收峰及β折叠结构,糖肽键类型为O-糖肽键,糖苷键类型为吡喃型。AGP中、高剂量组能显著提高免疫低下小鼠的细胞免疫和非特异性免疫功能。结论AGP-Ⅲ有糖蛋白的特征结构,AGP在适宜剂量条件下可提高小鼠的免疫能力。
OBJECTIVE: To analyze the primary structure of glycoprotein AGP-Ⅲ in the abalone of abalone Haliotis discus hannai and to investigate the immunological activity of AGP in crude glycoprotein. Methods The primary analysis of AGP-Ⅲ was carried out by high performance liquid chromatography (HPLC), amino acid composition analysis, infrared spectroscopy and β-elimination reaction. The immunosuppressive model of mice was established to investigate the regulation effect of AGP at different dosages on immune function. Results The monosaccharide composition of AGP-Ⅲ was fucose, galactose, grapes, mannose, galactosamine, xylose, glucose and rhamnose. AGP-Ⅲ contains 17 kinds of amino acids, valine content is the highest, followed by glutamic acid, glycine. AGP-Ⅲ glycoproteins exist in the characteristic absorption peak and β-sheet structure, glycopeptide bond type is O-glycopeptide bond, glycosidic bond type is pyran type. AGP, high dose group can significantly improve the immunosuppressive mice immune and non-specific immune function. Conclusions AGP-Ⅲ has the characteristic structure of glycoprotein. AGP can improve the immunity of mice under the condition of suitable dosage.
