本研究中所用的诺氏疟原虫是以恒河猴保存的虫种,所用的两株恶性疟原虫(氯喹敏感株和抗氯喹株)是以夜猴保存的虫种。诺氏疟原虫的体外培养用基本简化培养基,而恶性疟原虫的体外培养基用改进的Harvard培养基。红细胞提取物的制备是在室温下,将猴血用733g离心10分钟,除去血浆、淡黄色外层和少量压积红细胞,将剩下的细胞放在-70℃快速冰冻,而后在室温下融解。经冰冻
Plasmodium knowlesi used in this study was a species preserved in rhesus macaques. The two P. falciparums used (chloroquine-sensitive and chloroquine-resistant) were deposited as night-monkeys. In vitro culture of Plasmodium falciparum used minimally-simplified medium, whereas Plasmodium falciparum in vitro medium was modified with Harvard medium. Erythrocyte extract was prepared by centrifugation of the monkey’s blood at 733 g for 10 minutes at room temperature to remove the plasma, light yellow outer layer and a small amount of compressed red blood cells.
