目的:观察桃红四物汤对兔自体皮片移植模型Notch4受体表达的影响。方法:将60只新西兰大白兔随机分为4组,即桃红四物汤组、阻断剂组、模型组、假手术组,每组15只。造模后分组干预,并于术后第2,5,8天三个时限,从每组随机选取5只兔,行空气栓塞处死,切取移植皮片吻合区皮片组织,苏木精-伊红染色后在光镜下计算血管密度,采用免疫组化法测定Notch4受体蛋白表达情况。结果:桃红四物汤组移植区皮片的新生血管密度明显高于模型组、假手术组及阻断剂组(P<0.05),差异有统计学意义。Notch4受体蛋白表达含量桃红四物汤组表达量高于模型组、假手术组及阻断剂组(P<0.05),差异有统计学意义。结论:桃红四物汤能上调组织中血管新生相关因子Notch4受体的表达,促进移植皮片成活。
Objective: To observe the effect of Taohong Siwu Decoction on the expression of Notch4 receptor in rabbit autologous skin graft model. Methods: Sixty New Zealand white rabbits were randomly divided into four groups: Taohong Siwu Decoction group, blocker group, model group and sham operation group, with 15 rats in each group. After modeling, the rats were randomly divided into three groups: 5, 5, and 8 days after surgery. Five rabbits were randomly selected and killed by air embolization. The blood vessel density was calculated by light microscopy after red staining, and the expression of Notch4 receptor protein was detected by immunohistochemistry. Results: The neovascular density of skin graft in Taohong Siwu Decoction group was significantly higher than that in model group, sham operation group and blocker group (P <0.05). The difference was statistically significant. The expression of Notch4 receptor protein in Taohongsiwutang group was higher than that in model group, sham operation group and blocker group (P <0.05), the difference was statistically significant. Conclusion: Taohong Siwu Tang can up-regulate the expression of Notch4 receptor, an angiogenic factor, and promote the survival of the transplanted skin graft.
