目的:以鸭乙肝动物为模型,探讨芒果苷抑制鸭乙型肝炎病毒(DHBV)感染的免疫分子机制。方法:采用逆转录-聚合酶链反应法(RT-PCR)检测芒果苷对鸭脾细胞内IFN-γ、TNF-α、IL-2、IL-18等细胞因子(CK)mRNA表达的影响。结果:芒果苷高剂量组(200mg/kg)可增加鸭脾细胞中IL-18、IL-2、IFN-γ和TNF-αmRNA表达水平(P<0.05),并且两两之间的表达水平呈正相关性;芒果苷中剂量组(100mg/kg)可增强IL-18和IFN-γmRNA表达水平(P<0.05),对IL-2和TNF-αmRNA表达水平有上调的趋势(P>0.05);芒果苷低剂量组(50mg/kg)对IL-18、IL-2、IFN-γ和TNF-αmRNA表达水平无影响。结论:芒果苷可以提高脾细胞中IL-18、IL-2、IFN-γ和TNF-α等CKmRNA的表达水平,增强机体的Th1细胞功能,推测芒果苷可能通过增强机体的细胞免疫功能达到抗DHBV的作用。
OBJECTIVE: To investigate the immunological molecular mechanism of mangiferin in inhibiting duck hepatitis B virus (DHBV) infection using duck hepatitis B virus as a model. Methods: The effects of mangiferin on the mRNA expression of IFN-γ, TNF-α, IL-2 and IL-18 in duck spleen cells were detected by reverse transcription polymerase chain reaction (RT-PCR) RESULTS: High-dose mangiferin (200 mg / kg) increased the expression of IL-18, IL-2, IFN-γ and TNF-α mRNA in duck spleen cells (P <0.05) (100 mg / kg) could up-regulate the expression of IL-18 and IFN-γmRNA (P <0.05) and upregulate the expression of IL-2 and TNF-α mRNA (P> 0.05) The low dose of mangiferin (50mg / kg) had no effect on the expression of IL-18, IL-2, IFN-|Ã and TNF-|ÁmRNA. CONCLUSION: Mangiferin can increase the expression of CK mRNA such as IL-18, IL-2, IFN-γ and TNF-α and enhance the function of Th1 cells in spleen cells. It is speculated that mangiferin may enhance the cellular immune function of the body The role of DHBV.
