目的:考察以桑树为寄主不同季节的桑寄生总黄酮含量。方法:采用分光光度法测定以桑树为寄主不同季节12批桑寄生样品的总黄酮含量,并用正交实验的方法研究比较了不同提取条件对桑寄生总黄酮提取率的影响。结果:以桑树为寄主不同季节的桑寄生茎枝的总黄酮含量为7.63~7.86mg/g,叶总黄酮含量为31.52~32.08mg/g,其中以8月份桑寄生茎叶的总黄酮含量最高。正交试验结果优选的提取条件茎枝是A1B2C2D3,即桑寄生茎枝0.5g,乙醇浓度为60%,超声提取时间为30min,超声提取温度为45℃,料液比1∶25,叶是A2B1C2D3,即桑寄生叶0.5g,乙醇浓度为70%,超声提取时间为20min,超声提取温度为45℃,料液比1∶25。结论:桑树为寄主不同季节的桑寄生总黄酮含量因季节有所不同,其总黄酮含量表现出一定的差异性,但差异不大,桑寄生药材应全年可采。优选的提取方法稳定、可靠、简捷,提取效率高。
Objective: To investigate the content of total flavonoids in mulberry parasitic stems with mulberry as host in different seasons. Methods: The content of total flavonoids in 12 batches of mulberry parasitic plants sampled from mulberry trees in different seasons were determined by spectrophotometry. The effects of different extraction conditions on the extraction rate of total flavonoids from mulberry were studied by orthogonal experiment. Results: The content of total flavonoids in stems and leaves of mulberry parasitized by mulberry trees was 7.63-7.86mg / g and the content of total flavonoids in leaves was 31.52-32.08mg / g in different seasons. Among them, the content of total flavonoids in stems and leaves of mulberry leaves was the highest in August . Orthogonal test results are the best conditions for the extraction of stems A1B2C2D3, Sang parasitic stems 0.5g, ethanol concentration of 60%, ultrasonic extraction time of 30min, ultrasonic extraction temperature of 45 ℃, the ratio of material to liquid 1:25, leaves A2B1C2D3 , That is, mulberry leaf 0.5g, ethanol concentration of 70%, ultrasonic extraction time was 20min, ultrasonic extraction temperature was 45 ℃, the ratio of material to liquid 1:25. CONCLUSION: The content of total flavonoids of mulberry parasitism in different season of mulberry is different due to the different seasons. The content of total flavonoids shows a certain difference, but the difference is not obvious. The preferred extraction method is stable, reliable, simple and efficient.
