目的:山葛降脂分散片和葛根分散片中葛根素在大鼠体内药代动力学的对比研究。方法:以香草醛为内标物,利用HPLC测定大鼠血浆中葛根素的含量。结果:山葛降脂分散片的主要药动学参数:Tmax(3.000±0.371)h,Cmax(3.159±0.415)mg·L-1,AUC0→∞(19.593±3.168)mg·L-1·h-1,MRT0→∞(11.370±2.358)h;葛根分散片的主要药动学参数:Tmax(4.000±0.313)h,Cmax(2.784±0.398)mg·L-1,AUC0→∞(17.605±3.993)mg·L-1·h-1,MRT0→∞(8.678±1.625)h。结论:山楂叶和葛根提取物合用能使葛根素的吸收加快,浓度增大,消除减慢。房室模型拟合分析为二房室模型。
OBJECTIVE: To compare the pharmacokinetics of puerarin extracted from shangejiangzhi dispersible tablet and pueraria disperse tablet in rats. Methods: The content of puerarin in rat plasma was determined by HPLC with vanillin as internal standard. Results: The main pharmacokinetic parameters of Shanjiang Jiangzhi Dispersible Tablets: Tmax(3.000±0.371)h, Cmax(3.159±0.415)mg·L-1, AUC0→∞(19.593±3.168)mg·L-1·h -1,MRT0→∞(11.370±2.358)h; The main pharmacokinetic parameters of pueraria disperse tablet: Tmax(4.000±0.313)h, Cmax(2.784±0.398)mg·L-1, AUC0→∞(17.605±3.993) ) mg·L-1·h-1, MRT0→∞ (8.678±1.625) h. Conclusion: The combination of hawthorn leaves and Pueraria lobata extract can accelerate the absorption of puerarin, increase the concentration and eliminate the slow down. The room model fitting analysis was a two compartment model.
