目的观察乙肝转阴散(YGZYS)对鸭乙型肝炎病毒乙型肝炎表面抗原(DHBsAg)和乙型肝炎E抗原(DHBeAg)的抑制作用。方法采用1d龄雏鸭接种广西麻鸭乙型肝炎病毒(DHBV)强阳性血清,接种1周后用聚合酶链式反应(PCR)法筛选出DHBV强阳性鸭。随机分成5组:模型组、阳性对照组和YGZYS高、中、低剂量组,每组10只,各组雏鸭均灌胃给药14d。于用药前(T0)、用药7d(T7)和14d(T14)及停药后3d(P3)分别采血,采用酶联免疫吸附(ELISA)法测定血清DHBsAg和DHBeAg的滴度。结果与模型组比较,YGZYS高、中剂量组用药后血清DHBsAg和DHBeAg的滴度显著降低(P<0.01或P<0.05);停药3d后,YGZYS高、中剂量组血清DHBsAg和DHBeAg的滴度无反跳现象。YGZYS抑制DHBsAg和DHBeAg的作用有明显的量效和时效反应关系。结论 YGZYS可有效地抑制DHBsAg和DHBeAg的表达。
Objective To observe the inhibitory effect of YGZYS on duck hepatitis B virus surface antigen (DHBsAg) and hepatitis B E antigen (DHBeAg). Methods One-week-old ducklings were inoculated with strong positive sera of hepatitis B virus (DHBV) in Guangxi ducks. One week after inoculation, DHBV strongly positive ducks were screened by polymerase chain reaction (PCR). Randomly divided into 5 groups: model group, positive control group and YGZYS high, medium and low dose groups, each group of 10, each group of ducks were intragastrically administered 14d. Blood samples were collected before treatment (T0), 7d (T7) and 14d (T14) and 3d (P3) after drug withdrawal. The titer of serum DHBsAg and DHBeAg were determined by enzyme-linked immunosorbent assay (ELISA) Results Compared with the model group, the titer of serum DHBsAg and DHBeAg in YGZYS high and middle dose groups were significantly decreased (P <0.01 or P <0.05). After 3 days’ withdrawal, the serum levels of DHBsAg and DHBeAg No rebound phenomenon. YGZYS inhibition of the role of DHBsAg and DHBeAg significant dose-effect and time-response relationship. Conclusion YGZYS can effectively inhibit the expression of DHBsAg and DHBeAg.
