目的:比较长春碱(vinblastine,VLB)纳米粒(nanoparticles,NPS)和VLB生理盐水溶液对神经胶质瘤细胞C6生长和凋亡的影响。方法:体外培养的神经胶质瘤细胞C6分别用500μg.L-1的VLB NPS和VLB生理盐水溶液孵育,通过测定7 d内的细胞数量和2周后细胞的克隆形成能力,比较VLB NPS和VLB生理盐水溶液对神经胶质瘤细胞C6生长的抑制作用;在药物孵育48 h后,通过普通倒置显微镜和倒置荧光显微镜观察VLB NPS和VLB生理盐水溶液对神经胶质瘤细胞C6形态的影响,比较二者诱导神经胶质瘤细胞C6凋亡的能力。结果:VLB NPS与相同剂量的VLB生理盐水溶液相比,2~7 d时细胞数量显著下降;VLB生理盐水溶液组细胞的克隆形成率是VLB NPS溶液组的1.3倍,差异显著(P<0.05);形态学观察表明,VLB NPS溶液组的凋亡细胞数量明显多于VLB生理盐水溶液组,大部分细胞处于凋亡中、末期,失去结构完整性。结论:VLB NPS比相同剂量下的VLB生理盐水溶液抑制神经胶质瘤细胞C6生长和诱导其凋亡的能力更显著。
OBJECTIVE: To compare the effects of vinblastine (VLB) nanoparticles and VLB saline solution on glioma cell C6 growth and apoptosis. Methods: C6 glioma cells cultured in vitro were incubated with 500μg.L-1 VLB NPS and VLB saline respectively. By measuring the number of cells within 7 days and the clonogenic capacity of cells after 2 weeks, VLB NPS and VLB saline solution on glioma cell C6 growth inhibition; 48 h after the drug incubation, by ordinary inverted microscope and inverted fluorescence microscope VLB NPS and VLB saline solution on C6 glioma cell morphology, The ability to induce C6 glioma cell apoptosis was compared. Results: The number of cells in VLB NPS group was significantly decreased from 2 to 7 days when compared with the same dose of VLB saline solution. The rate of clone formation in VLB saline group was 1.3 times that of VLB NPS group (P <0.05) ). Morphological observation showed that the number of apoptotic cells in the VLB NPS solution group was significantly more than that in the VLB saline solution group, and most of the cells were in the middle and late stages of apoptosis, losing the structural integrity. CONCLUSION: VLB NPS has more pronounced inhibitory effect on glioma cell C6 growth and induction of apoptosis than the same dose of VLB saline solution.
