目的探讨碳酸酐酶Ⅸ(CA-Ⅸ)及缺氧诱导因子-1α(HIF-1α)在前列腺癌不同分期分级中的表达及其内在联系情况。方法采用免疫组织化学S-P法及Western-blot检测正常前列腺组织、前列腺癌组织以及前列腺癌细胞系PC-3、Lncap中CA-Ⅸ及HIF-1α的表达情况,结合临床资料进行统计分析,评价CA-Ⅸ及HIF-1α表达情况与前列腺组织癌变分化程度之间的关系,同时分析两者之间的相关性。结果在正常前列腺组织中CA-Ⅸ及HIF-1α基本不表达,在前列腺癌组织石蜡切片中,HIF处于高表达,其表达情况与前列腺癌病理分级相关。低分化的前列腺癌组织中HIF-1α的表达量高于高分化的前列腺癌组织。CA-Ⅸ在前列腺癌组织中表达率为37.5%,高于正常组织,与肿瘤分化程度无关。CA-Ⅸ及HIF-1α在前列腺癌组织中的表达情况具有相关性。结论 CA-Ⅸ及HIF-1α与前列腺癌的发生成正相关,而且两者在前列腺癌组织中的表达具有相关性,同时提示了以缺氧诱导因子通路为基础的分子机制在前列腺癌的演进中起到一定的作用。
Objective To investigate the expression of carbonic anhydrase Ⅸ (CA-Ⅸ) and hypoxia-inducible factor-1α (HIF-1α) in different stages of prostate cancer and its internal relations. Methods The expressions of CA-Ⅸ and HIF-1α in normal prostate tissue, prostate cancer and prostate cancer cell lines PC-3 and Lncap were detected by immunohistochemical SP method and Western-blot. The clinical data were used for statistical analysis and CA -IX and HIF-1αexpression and the degree of cancerous differentiation in prostate tissue, and analyze the correlation between the two. Results There was no expression of CA-Ⅸ and HIF-1α in normal prostatic tissues. HIF was highly expressed in paraffin-embedded sections of prostate cancer tissues, and its expression was correlated with the pathological grade of prostate cancer. The expression of HIF-1α in poorly differentiated prostate cancer is higher than that in well-differentiated prostate cancer. The expression of CA-Ⅸ was 37.5% in prostate cancer tissues, which was higher than that in normal tissues, which was not related to the degree of tumor differentiation. CA-Ⅸ and HIF-1α expression in prostate cancer has a correlation. Conclusions CA-Ⅸ and HIF-1α are positively correlated with the occurrence of prostate cancer, and their expression in prostate cancer is correlated with each other. At the same time, it is suggested that the molecular mechanism based on hypoxia-inducible factor pathway plays an important role in the progression of prostate cancer Play a role.
