目的:探讨五味子水提取液(SWE)对APP/PS1双转基因小鼠大脑皮层的神经细胞保护作用。方法:将C57BL/6小鼠分为正常组(Control),模型组(APP/PS1双转基因小鼠,Tg)和给药组(Tg+10、30、50 mg/kg的SWE,灌胃40 d);水迷宫检测各组小鼠的学习与记忆能力;Nissl法检测各组小鼠大脑皮层神经细胞的形态;TUNEL法检测各组小鼠大脑皮层神经细胞的凋亡情况;硫代巴比妥酸检测丙二醛(DMA)含量;黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)活性。结果:与模型组比较,灌胃SWE明显提高Tg小鼠的学习与记忆能力;改善大脑皮层神经细胞的形态;减少神经细胞凋亡;显著降低MDA的含量以及增强SOD的活性。结论:SWE对Tg小鼠大脑皮层的神经细胞有一定的保护作用,可能是通过减轻氧化应激造成的损伤来发挥治疗作用。
Objective: To investigate the neuroprotective effects of Schisandra chinensis fruit extract (SWE) on the cerebral cortex of APP / PS1 double transgenic mice. Methods: C57BL / 6 mice were divided into control group, model group (APP / PS1 double transgenic mice, Tg) and administration group (SWE with Tg + 10, 30 and 50 mg / kg) d). The learning and memory ability of mice in each group was detected by water maze. The morphology of neurons in cerebral cortex of each group was detected by Nissl method. The apoptosis of neurons in cerebral cortex of each group was detected by TUNEL method. The content of malondialdehyde (DMA) was determined by tocopherol acid test and the activity of superoxide dismutase (SOD) by xanthine oxidase method. Results: Compared with the model group, gavage of SWE significantly enhanced the learning and memory ability of Tg mice; improved the morphology of cerebral cortex neurons; decreased the apoptosis of neurons; significantly reduced the content of MDA and enhanced the activity of SOD. CONCLUSIONS: SWE has a protective effect on neural cells in Tg mouse cerebral cortex, which may play a therapeutic role by reducing the damage caused by oxidative stress.
