本研究应用农杆菌介导的方法,采用L_9(3~4)正交试验设计,对影响宁阳大枣基因转化的各项因子进行了分析比较,并着重探索了Ca~(2+)浓度对侵染转化的影响。结果表明:侵染效率最高的组合为1/2侵染液浓度,20 min侵染时间和0 mg/L Ca~(2+)浓度。Ca~(2+)浓度在提高宁阳大枣基因转化效率方面没有明显促进效果。Ca~(2+)浓度仅对平均分化芽数影响较大,浓度为0 mg/L时,平均分化芽数最多。本试验共获得27株转化苗,试验转化效率为33.33%。
In this study, Agrobacterium-mediated method was used to analyze and compare the factors affecting the gene transformation of jujube in Ningyang by using L_9 (3 ~ 4) orthogonal design.
