目的探讨慢性砷染毒对大鼠附睾上皮细胞凋亡的影响。方法将40只健康清洁雄性SD大鼠随机分为4组,分别为对照(蒸馏水)组和2.4、12.0、60.0 mg/L亚砷酸钠染毒组,每组10只。采用自由饮用方式进行连续染毒6个月。采用TUNEL细胞凋亡染色法测定大鼠附睾上皮凋亡细胞的凋亡情况,采用Western blot法测定附睾组织内caspase-3、caspase-9蛋白的表达水平,并测定精子顶体完整率和精子畸形率。结果与对照组比较,各浓度砷染毒组大鼠附睾管的细胞凋亡率较高,差异均有统计学意义(P<0.05);且随着砷染毒浓度的升高,大鼠附睾管的细胞凋亡程度加重。与对照组比较,各浓度砷染毒组大鼠附睾内caspase-3、caspase-9蛋白表达水平均较高,差异有统计学意义(P<0.05);且随着砷染毒浓度的升高,大鼠附睾内caspase-3、caspase-9蛋白的表达水平呈上升趋势。与对照组比较,12.0、60.0 mg/L砷染毒组大鼠精子顶体完整率均较低,而精子畸形率均较高,差异均有统计学意义(P<0.05);且随着砷染毒浓度的升高,大鼠精子顶体完整率呈下降趋势,而精子畸形率呈上升趋势。结论慢性砷染毒通过激活caspase-3或caspase-9等途径实现大鼠附睾组织的损伤及凋亡。
Objective To investigate the effects of chronic arsenic exposure on epididymal epithelial cell apoptosis in rats. Methods Forty healthy male Sprague-Dawley rats were randomly divided into 4 groups: control (distilled water) group and 2.4, 12.0, 60.0 mg / L sodium arsenite exposure group, 10 rats in each group. Continuous use of free drinking for 6 months. The apoptosis of epididymal epithelial cells was detected by TUNEL staining. The expression of caspase-3 and caspase-9 protein in epididymal epithelium was detected by Western blot. The rate of sperm acrosome integrity and sperm deformity rate. Results Compared with the control group, the apoptosis rates of the epididymal tubules of arsenic-treated rats at various concentrations were significantly higher (P <0.05). With the increase of arsenic concentration, The degree of apoptosis of the tube is aggravated. Compared with the control group, the expression levels of caspase-3 and caspase-9 in the epididymis group were higher than those in the control group (P <0.05). With the increase of arsenic concentration , The rat epididymal caspase-3, caspase-9 protein expression level is on the rise. Compared with the control group, the sperm acrosome intact rate of 12.0 and 60.0 mg / L arsenic-exposed rats was lower, while the sperm deformity rate was higher, the differences were statistically significant (P <0.05) Exposure to elevated concentrations, rat sperm acrosome intact rate showed a downward trend, while sperm deformity showed an upward trend. Conclusion Chronic arsenic exposure can induce epididymal injury and apoptosis in rats by activating caspase-3 or caspase-9.
