目的:建立在线固相萃取(Online SPE)结合HPLC测定复方甘草片中吗啡含量的方法。方法:采用Thermo Fisher Ultimate 3000双三元梯度液相色谱,以Acclaim C18(4.3 mm×10 mm,5μm)小柱对样品进行前处理和在线富集,后转载到分析柱Acclaim C18(4.6 mm×250 mm,5μm)上进行分离检测,流速为1.0 mL·min-1,检测波长220 nm。结果:吗啡在0.0239~0.239 mg·mL-1(r=0.999 9)范围内线性关系良好,平均加样回收率为99.19%。结论:该方法简便快捷,重现性好,节省大量时间与成本,方法精密度和准确度都有所提高,可为复方甘草片的质量控制提供方法。
OBJECTIVE: To establish a method for the determination of morphine in Compound Liquorice Tablets by on-line solid-phase extraction (SPE) combined with HPLC. METHODS: Samples were preconditioned and concentrated online on a Acclaim C18 (4.3 mm × 10 mm, 5 μm) column using a Thermo Fisher Ultimate 3000 dual ternary gradient LC and loaded onto an analytical column Acclaim C18 (4.6 mm × 250 mm, 5 μm). The flow rate was 1.0 mL · min-1 and the detection wavelength was 220 nm. RESULTS: Morphine had a good linearity in the range of 0.0239-0.239 mg · mL-1 (r = 0.999 9) with an average recovery rate of 99.19%. Conclusion: The method is simple and rapid, reproducible, and saves a lot of time and cost. The precision and accuracy of the method are improved. It can provide a method for the quality control of compound licorice tablets.
