采用RT-PCR和RACE-PCR的方法,从沙田柚果实中分离得到1个CgPIP2基因全长序列,编码284个氨基酸,与其它物种质膜水通道蛋白氨基酸序列比较结果表明,它们之间具有较高的同源性,且均具有水通道蛋白家族蛋白的特征结构单元NPA。采用烘干法测定贮藏期间沙田柚果实含水量的变化。结果表明:在沙田柚果实贮藏过程中,果皮和果肉的含水量逐步下降,且单果包装对果实含水量有一定的影响。半定量RT-PCR分析表明,CgPIP2在沙田柚果实的果皮和果肉中都有表达,随着贮藏期的延长表达逐渐减弱,但在果肉中的表达量稍高于果皮;另外,在贮藏20、40 d时,CgPIP2在单果包装沙田柚果实的果皮和果肉中的表达强于未经包装沙田柚果实果皮和果肉中的表达。因此,CgPIP2的表达可能与沙田柚贮藏过程中的水分含量变化有关。
A full-length CgPIP2 gene was isolated from the fruit of Shatian pomelo by RT-PCR and RACE-PCR, and encoded 284 amino acids. Therefore, the expression of CgPIP2 may be related to the change of water content during the storage of Shatian pomelo.
