尽管已知prohibitin1(PHB)与肿瘤有关,但很少有关于PHB在肝癌中的作用的报道.前期的糖蛋白质组学研究显示,PHB在高转移肝癌细胞株MHCC97-H细胞中被上调,且它可以被麦胚凝集素WGA所吸附.在本研究中,通过Western blotting和逆转录PCR实验证实,PHB在MHCC97-H细胞中的表达被上调约2倍.当PHB在MHCC97-H细胞中被过表达时,细胞增殖被抑制35%,细胞迁移率提高约2倍.本研究结果表明,PHB在MHCC97-H细胞中被上调,而且这种上调与肝癌细胞的增殖和迁移有关.
Although prohibitin1 (PHB) is known to be tumor related, few reports have been made on the role of PHB in hepatocellular carcinoma.Previous glycoproteomics studies showed that PHB is upregulated in highly metastatic hepatocellular carcinoma cell lines MHCC97-H and It can be adsorbed by wheat germ agglutinin WGA.In this study, the expression of PHB was up-regulated by about 2-fold in MHCC97-H cells by Western blotting and reverse transcription PCR experiments.When PHB was expressed in MHCC97-H cells The results showed that PHB was upregulated in MHCC97-H cells, and this up-regulation was related to the proliferation and migration of hepatoma cells.
