目的探讨甲醛对小鼠胚胎肢芽和中脑细胞分化、增殖的影响。方法分离12d龄小鼠胚胎肢芽和中脑细胞,与不同浓度的甲醛共培养,观察肢芽和中脑细胞的分化与增殖。结果随着甲醛染毒浓度的增加,胚胎肢芽细胞分化为软骨细胞的集落数逐渐减少,中脑细胞形成神经集落数减少、体积小,集落细胞间突起数量减少(P<0.01),肢芽细胞和中脑细胞增殖率下降(P<0.01),均具有剂量-效应关系;甲醛对肢芽细胞的半数分化抑制浓度(dID50)和半数增殖抑制浓度(pID50)分别为14.63μmol/L和27.67μmol/L,对中脑细胞的dID50和pID50分别为16.46μmol/L和26.92μmol/L。结论甲醛对胚胎中脑及肢芽细胞的分化和增殖有抑制作用,可能对胚胎骨和脑具有发育毒性。
Objective To investigate the effect of formaldehyde on the differentiation and proliferation of limb buds and midbrain cells in mouse embryos. Methods The limb buds and midbrain cells of 12-day-old mouse embryos were isolated and cultured with different concentrations of formaldehyde to observe the differentiation and proliferation of limb buds and midbrain cells. Results With the increase of the concentration of formaldehyde, the number of colony of embryo limb bud cells differentiated into chondrocytes decreased gradually, the number of colony of middle cerebral cells decreased, the volume of colony cells decreased, the number of colony cells decreased (P <0.01) (P <0.01), both of them had dose-effect relationship. The half-cell differentiation inhibitory concentration (dID50) and half-cell proliferation inhibitory concentration (pID50) of formaldehyde on limb bud cells were 14.63μmol / L and 27.67 μmol / L, respectively. The dID50 and pID50 of midbrain cells were 16.46μmol / L and 26.92μmol / L, respectively. Conclusion Formalin can inhibit the differentiation and proliferation of midbrain and limb bud cells in embryos, and may have developmental toxicity on embryonic bones and brain.
