目的了解大鼠热休克蛋白70(HSP70)的表达及肺组织病理改变与急性黄磷及其化合物吸入致急性肺损伤/急性呼吸窘迫综合征(ALI/ARDS)的关系。方法健康SD大鼠48只随机分为对照组以及造模后0、4、12、24、48h处死组,每组8只。采用自制的气体发生以及染毒装置,将大鼠置入染毒柜后,间歇染毒形成ALI/ARDS模型。观察ALI/ARDS大鼠动脉血气分析变化和肺系数,肺组织HSP70的表达以及肺组织病理改变。结果肺损伤后大鼠动脉血气以及肺组织病理改变明显恶化,肺系数较对照组明显增大,正常大鼠肺组织HSP70表达较弱,表达部位为肺泡上皮和部分支气管黏膜上皮细胞胞质;在ALI/ARDS条件下,大鼠肺组织HSP70表达逐渐增强,至成模后12h时相点,HSP70的表达最强,并呈现明显核表达;至成模后48h时相点,HSP70的表达逐渐减弱,核表达逐渐消失,主要在肺泡上皮和部分支气管黏膜上皮细胞胞质表达。结论急性黄磷及其化合物吸入致ALI/ARDS大鼠肺组织病理改变随造模后时相的延长越来越严重。ALI/ARDS条件下大鼠肺组织HSP70表达表现为:受到应激-高表达-表达减弱,并伴有HSP70的再分布。
Objective To investigate the relationship between the expression of heat shock protein 70 (HSP70) and the pathological changes of lung tissue and the acute lung injury / acute respiratory distress syndrome (ALI / ARDS) induced by acute inhalation of yellow phosphorus and its compounds. Methods Forty - eight healthy SD rats were randomly divided into control group and sacrificed at 0, 4, 12, 24, 48 h after model establishment, with 8 in each group. Using homemade gas generation and exposure devices, rats were exposed to an exposure cabinet and then intermittently exposed to ALI / ARDS model. Arterial blood gas analysis and lung index, lung tissue HSP70 expression and lung histopathological changes were observed in ALI / ARDS rats. Results After the lung injury, the arterial blood gas and the histopathological changes of the lung tissue were significantly deteriorated. The lung coefficient was significantly higher than that of the control group. The expression of HSP70 in the lung tissue of normal rats was weak, and the expression sites were cytoplasm of alveolar epithelium and some bronchial epithelial cells. Under the condition of ALI / ARDS, the expression of HSP70 in the lung tissue of rats gradually increased. The expression of HSP70 was the strongest at the point of 12h after modeling, and the expression of HSP70 was obvious. At the point of 48h, the expression of HSP70 gradually decreased , Nuclear expression gradually disappeared, mainly in the alveolar epithelium and some bronchial epithelial cells cytoplasmic expression. Conclusion The pathological changes of lung tissue in ALI / ARDS rats after acute yellow phosphorus and its inhalation are more and more serious with the prolongation of the phase after modeling. Under the condition of ALI / ARDS, the expression of HSP70 in the lungs of rats showed that the expression of HSP70 was decreased due to the stress-high expression-accompanied with the redistribution of HSP70.
