目的:观察地菍口服液对原发免疫性血小板减少症(primary immune thrombocytopenia,ITP)模型小鼠血小板计数、出凝血时间、骨髓巨核细胞数量、血清白细胞介素-4(IL-4)及干扰素-γ(IFN-γ)的影响。方法:取60只BALB/C小鼠随机分为6组:地菍口服液高、中、低剂量组、醋酸泼尼松组、模型组、正常对照组。以隔日注射豚鼠抗小鼠血小板血清(APS)进行造模。造模第8天开始给药,每日1次,连续14 d后处死小鼠,检测外周血小板计数、出凝血时间、骨髓巨核细胞数量、IL-4及IFN-γ水平。结果:1地菍口服液高、中、低剂量组血小板数量明显高于模型组(P<0.05),出血时间及巨核细胞计数明显低于模型组(P<0.05);地菍口服液高剂量组凝血时间低于模型组(P<0.05),但中、低剂量与模型组间比较无显著变化(P>0.05);地菍口服液高、中剂量组血小板计数高于低剂量组(P<0.05)。2模型组IL-4、IFN-γ及IFN-γ/IL-4比值水平均高于正常对照组(P<0.05),以IFN-γ及IFN-γ/IL-4值升高为主(P<0.05);地菍口服液高剂量组IL-4、IFN-γ水平及IFN-γ/IL-4值低于模型组(P<0.05),而中、低剂量组仅IFN-γ水平及IFN-γ/IL-4值低于模型组(P<0.05);地菍口服液高、中、低剂量组IL-4、IFN-γ及IFN-γ/IL-4值无明显差异(P>0.05)。结论:地菍口服液对于提升ITP小鼠血小板计数及改善出血症状有较好疗效,增加剂量有助于提高疗效。通过降低ITP小鼠INF-γ水平及IFN-γ/IL-4比值,使Th1/Th2细胞恢复平衡,可能是其起效的机制之一。
Objective: To observe the effect of Di-tong oral liquid on platelet count, clotting time, the number of bone marrow megakaryocytes, serum interleukin-4 (IL-4) and interference in primary immune thrombocytopenia (ITP) Effects of Superolein-γ (IFN-γ). Methods: Sixty BALB / C mice were randomly divided into 6 groups: Dihuang Oral Liquid high, medium and low dose groups, prednisone acetate group, model group and normal control group. Guinea pig anti-mouse platelet serum (APS) was injected every other day for modeling. The mice were sacrificed on the 8th day after the operation, once a day for 14 days. The peripheral blood platelet count, the clotting time, the number of the bone marrow megakaryocytes, the levels of IL-4 and IFN-γ were measured. Results: The numbers of platelets in high, middle and low doses of Dihuang oral liquid group were significantly higher than those in model group (P <0.05), and the bleeding time and megakaryocyte count were significantly lower than those in model group (P <0.05) (P <0.05). However, there was no significant difference between middle and low dose groups and the model group (P> 0.05). The platelet count of high and medium dose Di Di oral liquid group was higher than that of low dose group <0.05). 2 and IL-4, IFN-γ and IFN-γ / IL-4 in the model group were significantly higher than those in the normal control group (P <0.05) (P <0.05). The levels of IL-4, IFN-γ and IFN-γ / IL-4 in SD group were lower than those in DM group (P <0.05) (P <0.05). There was no significant difference in the levels of IL-4, IFN-γ and IFN-γ / IL-4 among the DM, DM and low dose groups P> 0.05). Conclusion: Diji Oral Liquid can improve the platelet count and improve the bleeding symptoms of ITP mice. Increasing the dose of Dihuang Oral Liquid can improve the curative effect. It may be one of the mechanisms of its effect by restoring the balance of Th1 / Th2 cells by decreasing the level of INF-γ and the ratio of IFN-γ / IL-4 in ITP mice.
