[目的]对鸡importin β1基因进行克隆和生物信息学分析。[方法]根据Gen Bank公布的鸡importin β1基因序列(XM_015299473)设计合成特异性引物,通过RT-PCR从DF-1细胞中扩增importin β1基因CDS区并进行克隆和测序,利用生物信息学工具对其理化性质、二级结构和系统进化树进行分析。[结果]鸡importin β1基因CDS全长2 268bp,共编码755个氨基酸,该蛋白等电点为4.61,相对分子质量84.15 k Da,分子式为C3712H5901N979O1152S46。蛋白质二级结构预测显示,鸡importin β1蛋白含有丰富的二级结构,以α-螺旋为主(占64.90%)。importin β1基因同源性及系统进化树分析结果表明鸡与鹌鹑的亲缘关系最近。[结论]鸡importin β1基因的克隆和生物信息学分析为进一步研究其功能奠定了基础。
[Objective] The research aimed to clone and bioinformatically analyze the importin β1 gene of chicken. [Method] The specific primers were designed and synthesized according to the sequence of chicken importin β1 gene (GenBank accession number: XM_015299473) published by Gen Bank. The CDS region of importin β1 gene was amplified from DF-1 cells by RT-PCR and cloned and sequenced. Bioinformatics tools Its physical and chemical properties, secondary structure and phylogenetic tree analysis. [Result] The full - length CDS of importi β1 gene of chicken was 2 268 bp, encoding a total of 755 amino acids. Its isoelectric point was 4.61, relative molecular mass was 84.15 kDa and molecular formula was C3712H5901N979O1152S46. Protein secondary structure prediction showed that the chicken importin β1 protein is rich in secondary structure, mainly α-helix (64.90%). The results of phylogenetic tree analysis and phylogenetic tree analysis showed that the genetic relationship between importin β1 gene and quail was the closest. [Conclusion] Cloning and bioinformatics analysis of importin β1 gene in chicken laid the foundation for further study of its function.
