目的探讨乙型肝炎病毒(HBV)相关肝癌患者血清中miR-205-5p的表达水平及其临床意义。方法收集34例HBV相关肝癌患者、41例慢性肝病患者、19例健康对照血清样本,采用实时荧光定量PCR(qRT-PCR)检测其miR-205-5p的表达水平,受试者工作特征(ROC)曲线分析血清miR-205-5p用于HBV相关肝癌诊断的价值。结果 HBV相关肝癌患者血清miR-205-5p表达水平低于健康对照组与慢性肝病组,差异有统计学意义(P<0.05)。不同血清甲胎蛋白水平、乙肝病毒载量、病灶数、肿瘤大小及癌栓的HBV相关肝癌患者之间,血清miR-205-5p表达无相关性(P>0.05)。ROC曲线分析显示,血清mi R-205-5p区分HBV相关肝癌患者与健康对照的ROC曲线下面积(AUC)为0.812,诊断敏感度与特异度分别为79.4%、73.7%。结论 miR-205-5p有望成为一种新的HBV相关肝癌诊断标记物。
Objective To investigate the expression of miR-205-5p in serum of hepatitis B virus (HBV) -related hepatocellular carcinoma patients and its clinical significance. Methods Serum samples from 34 patients with HBV-related hepatocellular carcinoma, 41 patients with chronic liver disease and 19 healthy controls were collected. The expression of miR-205-5p was detected by real-time quantitative PCR (qRT-PCR), and the receiver operating characteristic (ROC ) Curve analysis of serum miR-205-5p for the diagnosis of HBV-related liver cancer. Results The expression of miR-205-5p in HBV-related liver cancer patients was lower than that in healthy controls and chronic liver disease patients (P <0.05). There was no correlation between the expression of miR-205-5p and the HBV-related hepatocellular carcinoma patients with different levels of serum alpha-fetoprotein, hepatitis B virus load, number of lesions, tumor size and tumor embolus (P> 0.05). ROC curve analysis showed that the area under the ROC curve (AUC) of serum mi R-205-5p in separating HBV-related hepatocellular carcinoma from healthy controls was 0.812, and the diagnostic sensitivity and specificity were 79.4% and 73.7%, respectively. Conclusion miR-205-5p is expected to become a new diagnostic marker of HBV-related liver cancer.
