目的:探讨膀胱癌患者尿液中微小核糖核酸17-5p(miR-17-5p)表达及其诊断价值。方法运用荧光定量PCR检测miR-17-5p在膀胱癌患者尿液中的表达水平,采用△△CT方法计算膀胱癌患者与正常人尿液中miR-17-5p表达量的比值,结合术后病理参数,统计分析miR-17-5p与临床病理资料的相关性。结果膀胱癌患者尿液中miR-17-5p表达较正常人明显升高。26例膀胱癌患者中18例(69.2%)的miR-17-5p表达较正常人显著升高,且与肿瘤大小和恶性程度相关,肿瘤越大、恶性程度越高,尿液中miR-17-5p的表达量越高。结论尿液中高表达miR-17-5p可能成为诊断膀胱癌的重要标志物,并与膀胱癌的临床病理密切相关,可能对膀胱癌的诊断提供新的思路。“,”Objective Our purpose was to investigate the expression of microRNA-17-5p(miR-17-5p)in urine from the patients of bladder cancer. Methods Real-time RT-PCR was used to detect the expression of miR-17-5p in urine, whose data were analyzed by the relatively quantitative 2-△△CT method. Then, we analyzed the association between miR-17-5p expression and clinicopathological characteristics in bladder cancer by Chi-Square Test. Results The results showed that the expression of miR-17-5p increased significantly compared with that of the normal control. Among 26 patients, 18 (69.2%) cases showed an obvious high-expression of miR-17-5p. Meanwhile, there was a significant positive correlation between the expression of miR-17-5p and tumor size/malignant degree. Conclusion The high expression of miR-17-5p in urine might be involved in the tumorigenesis and development of bladder cancer, indicating its possibility of becoming an important marker for bladder cancer, and of providing a new diagnostic strategy.
