目的探讨c-Jun氨基末端激酶选择性抑制剂(SP600125)对哮喘小鼠肺组织白介素-2(IL-2)表达的影响。方法 30只BALB/c小鼠随机分为对照组、哮喘组和SP600125组,制作哮喘模型及干预处理后,处死小鼠,取肺组织,采用免疫组织化学方法和Western blot方法检测各组小鼠肺组织内IL-2的表达。结果免疫组化结果显示,哮喘组小鼠肺组织IL-2表达的平均光密度值为0.76±0.11,显著高于对照组0.25±0.04,P<0.01;而SP600125组小鼠肺组织IL-2表达的平均光密度值为0.45±0.08,与哮喘组相比明显降低,P<0.01。Western blot结果显示,哮喘组小鼠肺组织IL-2表达的平均光密度值为1.26±0.21,显著高于对照组0.36±0.05,P<0.01;而SP600125组小鼠肺组织IL-2表达的平均光密度值为0.88±0.11,与哮喘组相比明显降低,P<0.01。结论 c-Jun氨基末端激酶选择性抑制剂能降低哮喘小鼠肺组织IL-2的表达。
Objective To investigate the effect of c-Jun N-terminal kinase selective inhibitor (SP600125) on the expression of interleukin-2 (IL-2) in the lung tissue of asthmatic mice. Methods Thirty BALB / c mice were randomly divided into control group, asthma group and SP600125 group. After asthma model was established and intervention treatment was given, the mice were sacrificed and the lungs were removed. Immunohistochemistry and Western blot were used to detect the expression of IL-2 expression in lung tissue. Results Immunohistochemical results showed that the average optical density of IL-2 expression in the lung tissue of asthmatic mice was 0.76 ± 0.11, significantly higher than that of the control group (0.25 ± 0.04, P <0.01); while the SP600125 mice lung IL-2 The average optical density value was 0.45 ± 0.08, which was significantly lower than that in asthma group (P <0.01). Western blot results showed that the average optical density of IL-2 expression in lung tissue of asthmatic mice was 1.26 ± 0.21, which was significantly higher than that of control group (0.36 ± 0.05, P <0.01), while the expression of IL-2 in lung of SP600125 mice The average optical density was 0.88 ± 0.11, which was significantly lower than that in the asthma group (P <0.01).
