为考察硫熏及直接热风加热的粉葛中葛根素含量及药效的差异,将鲜粉葛削皮、切小块分别制备成晒干、直接热风干燥及硫熏后热风干燥样品,分别测定样品水分含量。采用HPLC,比较晒干、直接热风干燥及硫熏后热风干燥样品中粉葛中主要药效成分葛根素的含量;建立醉酒模型,以SOD含量为指标,灌胃给药硫熏和非硫熏粉葛样品的水煎煮提取物,比较硫熏对粉葛提高醉酒模型小鼠肝脏、血清中SOD含量的影响;同时考察不同包装及贮存条件下,硫熏及非硫熏样品的霉变及虫蛀情况。结果显示硫熏后的粉葛样品,其葛根素含量发生明显变化,随着硫熏次数及硫磺用量的提高,其葛根素含量逐渐下降。且硫熏可显著降低粉葛提高的醉酒模型小鼠肝脏、血清中SOD含量,与晒干及直接热风干燥组均呈显著性差异。水分符合药典标准的硫熏样品和直接热风干燥样品均未发生虫蛀和霉变。研究显示硫熏可显著降低粉葛中主要药效成分的含量,降低粉葛的药效,表明硫熏后粉葛质量显著下降。而未硫熏直接热风干燥的样品其主要药效成分的含量、药效以及贮藏结果均与晒干相似,是一种较好的产地干燥工艺,可推广。
