本试验采用生物素—亲和素酶复合物—斑点酶联免疫吸附试验(ABC—Dot—ELISA)检测食品中沙门氏菌。该法可检出ISC液中接种的沙门氏菌。其敏感性可达104—105千个/L,比常规免疫酶法和免疫荧光法提高10—100倍。特异性检测表明,其与大肠杆菌、变形杆菌、产气杆菌未发生交叉反应。该法在24h内可报告结果,比常规法至少快3—4d。对200份食品检样进行检测,检出19份,检出率9.5%,与常规法检测结果平行比较,漏检率仅为0.5%,结果相差不显著(P>0.05)。进行3次重复性试验,结果完全相同。试验结果表明,该法操作简便、敏感、快速,具有较高的特异性,适于对大批样品进行快速检测。
In this study, the detection of Salmonella in food by biotin-avidin complex-dot ELISA (ABC-Dot-ELISA). The method can detect ISC liquid inoculated Salmonella. The sensitivity of up to 104-105 thousand / L, than conventional immunoenzymatic and immunofluorescence method to improve 10-100 times. Specificity tests showed that it did not cross-react with Escherichia coli, Proteus, and Aerobacter aerogenes. The method can report the results within 24 hours, at least 3-4d faster than the conventional method. The detection rate of 200 food samples was 19 and the detection rate was 9.5%. The detection rate was only 0.5% compared with the conventional method. The difference was not significant (P> 0.05 ). Repeat the test three times, the results are exactly the same. The experimental results show that the method is simple, sensitive, rapid, and has high specificity and is suitable for rapid detection of a large number of samples.
