目的研究口腔鳞癌(OSCC)中miR-15b的表达及其对OSCC Cal27细胞增殖和凋亡能力的影响。方法实时定量PCR检测2013年1月至2015年1月期间中国医科大学附属口腔医院外科50例住院患者OSCC组织中mi R-15b基因的表达水平。培养OSCC Cal27细胞,将mi R-15b的前体转染Cal27细胞,MTT法及双标流式法检测转染前后Cal27细胞的增殖和凋亡能力。结果与癌旁组织相比,mi R-15b在OSCC组织中的表达显著下调,且与OSCC的分级相关。mi R-15b的前体能够使Cal27细胞的增殖能力明显下降,且能够诱导Cal27细胞发生凋亡。结论 mi R-15b的异常表达与OSCC相关,mi R-15b能够抑制OSCC细胞增殖并诱导其凋亡,在OSCC中发挥肿瘤抑制基因的作用。
Objective To investigate the expression of miR-15b in oral squamous cell carcinoma (OSCC) and its effect on the proliferation and apoptosis of OSCC Cal27 cells. Methods Real-time quantitative PCR was used to detect the expression of mi R-15b gene in OSCC tissue from 50 inpatients of Department of Stomatology, China Medical University from January 2013 to January 2015. The cells of OSCC Cal27 were cultured, and the precursor of mi R-15b was transfected into Cal27 cells. The proliferation and apoptosis of Cal27 cells before and after transfection were detected by MTT and double-labeled flow cytometry. Results The expression of mi R-15b was significantly downregulated in OSCC tissues compared with that in paracancerous tissues, and correlated with the grade of OSCC. The precursor of mi R-15b could significantly reduce the proliferation of Cal27 cells and induce the apoptosis of Cal27 cells. Conclusion The abnormal expression of mi R-15b is related to OSCC. Mi R-15b can inhibit the proliferation and induce the apoptosis of OSCC cells, and play a role of tumor suppressor gene in OSCC.
