目的:实验研究紫杉醇(PA)、白藜芦醇(Res)合用对人胃癌MGC803细胞影响。方法:通过流式细胞仪(FCM)检测细胞周期,Western blot。方法:研究PA与Res合用前后对体外培养的人胃癌细胞MGC803对细胞的影响。结果:FCM显示细胞生长停止在S期,S期数分数为0.52,抑制细胞有丝分裂及DNA合成。Res与PA合用可以下调Bcl-2表达,上调Bax表达,对P-gp作用不明显。结论:PA与小剂量化疗药物Res合用,可获得化疗药物单独用药相同疗效下调Bcl-2表达,上调Bax表达,增加敏感性,减少药物副作用。
OBJECTIVE: To study the effects of combination of paclitaxel (PA) and resveratrol (Res) on human gastric cancer MGC803 cells. Methods: The cell cycle was detected by flow cytometry (FCM) and Western blot. Methods: The effects of PA and Res on the cell proliferation of human gastric cancer cells MGC803 cultured in vitro were studied. RESULTS: FCM showed that the cell growth stopped at S phase and the S phase fraction was 0.52, which inhibited cell mitosis and DNA synthesis. The combination of Res and PA could down-regulate the expression of Bcl-2 and up-regulate the expression of Bax, but the effect of P-gp was not obvious. Conclusion: Combination of PA and low-dose Chemotherapy Res can reduce the expression of Bcl-2, increase the expression of Bax, increase the sensitivity and reduce the side effects of drugs.
