研究在磷酸缓冲介质(PBS)中,一种检测己烯雌酚抗原的新型免标记电化学免疫传感器的制备及应用。首先将纳米金(AuNP)修饰在电极上,然后将石墨烯(Gr)-壳聚糖(CS)复合物饰于玻碳电极表面,通过循环伏安法对修饰的电极进行表征。以[Fe(CN)6]3-/4-为氧化-还原探针,基于己烯雌酚抗原抗体反应引起[Fe(CN)6]3-/4-探针的电流响应的变化,来实现对己烯雌酚抗原的检测。己烯雌酚抗原的质量浓度在0.5~1500.0ng/mL范围,与峰电流呈良好的线性关系,相关系数为0.985,检测限为0.1ng/mL。该传感器具有良好的重现性和稳定性,用于动物组织和奶粉样品的测定获得满意结果。
To study the preparation and application of a novel label-free electrochemical immunosensor for the detection of diethylstilbestrol antigens in phosphate buffered saline (PBS). Firstly, AuNP was modified on the electrode, and then the graphene (Gr) -chitosan (CS) composite was decorated on the surface of glassy carbon electrode. The modified electrode was characterized by cyclic voltammetry. Based on the change of the current response of [Fe (CN) 6] 3- / 4-probe based on the reaction of diethylstilbestrol antigen with [Fe (CN) 6] 3- / 4- as oxidation-reduction probe, Antigen detection. The concentration of diethylstilbestrol antigen in the range of 0.5 ~ 1500.0ng / mL, and the peak current was a good linear relationship, the correlation coefficient of 0.985, detection limit of 0.1ng / mL. The sensor has good reproducibility and stability, for animal tissue and milk powder samples obtained satisfactory results.
