目的 了解 3个 Y染色体上短串联重复序列 (short tandem repeat on Y chromosome,Y-STR)基因座在隔离群体中的多态性 ,探讨用 Y- STR分析群体亲缘关系的可能性。方法 在 377测序仪上应用基因扫描技术对浙江省两个海岛隔离群体 (80人和 6 0人 ) ,和 1个开放群体 (36人 )的 DYS388、DYS390、DYS395进行检测。结果 DYS388、DYS390、DYS395在渔山岛、桃花岛、开放群体中的等位基因数分别为 8、9、7;5、6、7;6、6、5。 3个群体中每个 Y- STR基因的基因多样性在 0 .70~ 0 .80之间。隔离群体和开放群体在等位基因的频率分布、相同基因型数的分布差异无显著性。遗传距离最大的为桃花岛与开放群体 ,其次是渔山岛与桃花岛群体 ,最小的是渔山岛和开放群体。结论 DYS388的常见等位基因为 12 9,DYS390、DYS395分别为 2 15和 119。 DYS388、DYS390、DYS395的基因频率、基因多样性、群体的相同基因型分布和遗传距离均不能说明隔离群体之间、以及与开放群体的亲缘关系程度 ,提示用 Y- STR分析群体亲缘关系须在增加样本量的基础上进行继续深入的研究。
Objective To understand the polymorphism of the short tandem repeat on Y chromosome (Y-STR) loci in three segregants in Y chromosome and to explore the possibility of using Y-STR to analyze the genetic relationship among populations. Methods The DYS388, DYS390 and DYS395 of two island isolates (80 and 60) and one open group (36) in Zhejiang Province were detected by gene scanning technique on a 377 sequencer. Results The numbers of alleles of DYS388, DYS390 and DYS395 in Yushan Island and Taohua Island were 8, 9, 7, 5, 6, 7, 6, 6 and 5 respectively. The genetic diversity of each Y-STR gene in three populations ranged from 0.70 to 0.80. There was no significant difference in the frequency distribution of alleles among isolates and open groups with the same number of genotypes. The largest genetic distance is Peach Blossom Island and open groups, followed by Yushan Island and Taohua Island groups, the youngest is Yushan Island and open groups. Conclusion The common allele of DYS388 is 129, DYS390 and DYS395 are 2 15 and 119, respectively. DYS388, DYS390, DYS395 gene frequency, gene diversity, the same population of the same genotype distribution and genetic distance can not explain the isolated groups, as well as the degree of genetic relationship with the open group, suggesting that using Y-STR analysis of genetic relationships in the group to be in Increase the sample size based on continuing in-depth research.
