目的:研究降脂膏(JZG)对高脂大鼠一氧化氮/内皮素、前列环素/血栓烷B_2及血液流变学的影响。方法:用高脂乳剂喂饲40d以形成大鼠高脂血症及动脉粥样硬化模型,同时给药组igJZG40、60、80/kg,测定血中NO、ET-1、6-keto-PGF_(1a)、TXB_2及血液流变学各指标。结果:JZG对正常及高脂大鼠血清NO水平有一定升高,明显降低ET-1水平,大剂量能显著升高、6-keto-PGF_(1a)含量,有利于维持6-keto-PGF_(1a)-PGF_1/ TXB_2平衡;可降低全血比粘度、全血还原比粘度、血浆比粘度、降低RBC聚集指数、压积,缩短RBC电泳时间,并能降低纤维蛋白原含量,对二磷酸腺苷诱导的正常大鼠血小板聚集,亦有一定的抑制作用。结论:JZG改善高脂大鼠血液流变学异常、恢复NO、ET-1、6-keto-PGF_(1a)、TXB_2平衡,可能是其抗动脉粥样硬化作用的机制之一。
Objective: To study the effects of Jiang Zhi Gao (JZG) on nitric oxide/endothelin, prostacyclin/thromboxane B_2 and hemorheology in rats with high fat. METHODS: Rats with hyperlipidemia and atherosclerosis were treated with high-fat emulsion for 40 days. At the same time, igJZG 40, 60, and 80/kg were administered to the rats to determine NO, ET-1, and 6-keto-PGF_ in blood. (1a), TXB_2, and hemorheology indexes. RESULTS: The level of NO in serum of normal and hyperlipid rats was increased by JZG, and the level of ET-1 was significantly decreased. Large doses could significantly increase the content of 6-keto-PGF_(1a), which was beneficial to the maintenance of 6-keto-PGF_. (1a)-PGF_1/TXB_2 balance; can reduce whole blood specific viscosity, whole blood reduction specific viscosity, plasma specific viscosity, decrease RBC aggregation index, and product volume, shorten RBC electrophoresis time, and can reduce fibrinogen content, for diphosphate Adenosine-induced platelet aggregation in normal rats also has a certain inhibitory effect. Conclusion: JZG can improve the abnormal hemorheology and restore the balance of NO, ET-1, 6-keto-PGF_(1a) and TXB_2 in hyperlipidemia rats, which may be one of the mechanisms of its anti-atherosclerosis effect.
