目的探索愤怒、郁怒情绪与γ氨基丁酸B2受体(GABABR2)的关系,以及调肝方药经前平颗粒和经前舒颗粒的中枢干预机制。方法采用居住入侵和社会隔离的方法复制愤怒、郁怒情绪大鼠模型,用RT-PCR和Western Blot的方法检测GABABR2 mRNA和蛋白的表达差异。结果与正常对照组相比,愤怒、郁怒情绪模型大鼠下丘脑GABABR2 mRNA水平和蛋白水平均明显下降(P<0.01~0.05),而且愤怒模型组的降低程度明显大于郁怒模型组。与各模型组相比,各给药组GABABR2 mRNA水平和蛋白水平均有不同程度的升高。结论大鼠下丘脑GABABR2表达降低可能是影响大鼠愤怒和郁怒情绪的重要共性机制之一;中药经前平和经前舒颗粒对GABABR2表达异常变化具有调节作用。
Objective To explore the relationship between the emotions of anger and emotions and γ-aminobutyric acid B2 receptor (GABABR2), and to investigate the mechanism of intervention of jianqian granule and Jingqianshu granule. Methods The living and social isolation methods were used to copy the model of anger and depression and anger, and the expression of GABABR2 mRNA and protein was detected by RT-PCR and Western Blot. Results Compared with the normal control group, the expression of GABABR2 mRNA and protein in the hypothalamus of the rats with anger and depression mood were significantly decreased (P <0.01 ~ 0.05), and the degree of depression in the anger model group was significantly greater than that of the depression model group. Compared with the model group, the GABABR2 mRNA and protein levels in all the groups were increased to some extent. Conclusion The decrease of GABABR2 expression in rat hypothalamus may be one of the most important common mechanisms that may affect the anger and depression of rats. Jingjing Hejing Jingqianshu Granule can regulate the abnormal expression of GABABR2 in rats.
