目的探讨Toll样受体4(TLR4)的表达与动脉粥样硬化(AS)及其主要危险因素的关系。方法选取行冠状动脉旁路移植术患者31例共63块主动脉壁全层组织作为研究组;肾移植供体者8名共32块动脉壁全层组织作为对照组。应用免疫组织化学方法检测动脉壁全层组织中TLR4蛋白的表达,并分析TLR4蛋白的表达与年龄、性别、吸烟、血压、血糖、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、总胆固醇(TC)、甘油三酯(TG)等AS危险因素之间的关系。结果TLR4在研究组动脉壁全层组织中有明显表达,而对照组无表达。TLR4表达与高血压、高血糖、LDL-C升高、HDL-C降低相关(P均<0.05),与年龄、性别、吸烟、TC、TG无相关性(P>0.05)。结论TLR4可能是AS的始动因素,高血压、高血糖、LDL-C升高、HDL-C降低等危险因素通过TLR4信号途径促进AS的发生发展。
Objective To investigate the relationship between the expression of Toll-like receptor 4 (TLR4) and atherosclerosis (AS) and its major risk factors. Methods A total of 63 aorta walls of 31 patients undergoing coronary artery bypass grafting were selected as the study group. Eight allograft renal artery donors were used as the control group. Immunohistochemistry was used to detect the expression of TLR4 protein in the whole layer of arterial wall. The expression of TLR4 protein was analyzed with age, gender, smoking, blood pressure, blood glucose, high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), total cholesterol (TC), triglyceride (TG) and other AS risk factors. Results TLR4 was obviously expressed in the whole artery of the study group but not in the control group. The expression of TLR4 was associated with hypertension, hyperglycemia, elevated LDL-C, and decreased HDL-C (all P <0.05), but not with age, gender, smoking, TC, TG. Conclusion TLR4 may be the initiating factor of AS. Risk factors such as hypertension, hyperglycemia, elevated LDL-C and decreased HDL-C may promote the development of AS through TLR4 signaling pathway.
