母种培养基,一般都用琼脂作凝固剂,但在山区农村难以买到琼脂。为此,我进行了大米磨浆代琼脂试验,取得了较好效果,现将具体做法简介如下:将大米用温水浸泡12小时,加水量为超过米面2~3厘米。浸后用磨磨成浓浆(反复磨2~3次),然后加适量葡萄糖、磷酸二氢钾,拌匀后分装试管,装量为试管长的1/5,然后取一个平底茶盆,盆内放一条小木棒把试管搁在木棒上摆成斜面,置洁净的铁锅蒸架上(离水面2~3厘米),用猛火蒸至冒气后维持30~45分钟起锅,此时培养基已凝结成半透明的斜面。如果试管内有水珠,要倒掉水后再塞棉塞、包扎牛皮纸,进行高压灭菌。
