目的:观察补肾活血中药对大鼠慢性高眼压(elevated intraocular pressure,EIOP)模型初级视皮质尼氏小体损害的干预作用,并对其作用机理进行初步探讨。方法:采用烙闭上巩膜静脉法,烙闭大鼠3支上巩膜静脉,制作大鼠慢性EIOP模型,随机分为3组:空白组,模型组,给药组。连续灌胃8wk,并于8wk末处死大鼠,观察其对EIOP大鼠眼压(intraocular pressure,IOP),初级视皮质(primary visual cortex,PVC)尼氏小体含量、神经元细胞超微结构影响。结果:本实验采用的烙闭上巩膜静脉的造模方法使大鼠眼压明显升高(P<0.01),与造模前比较差异有显著统计学意义(P<0.01);PVC病理切片半定量分析表明:模型组尼氏小体总面积34941±8482.1·S/μm2、平均光密度152.8±27.97、积分光密度11993±3084.8,与空白组(总面积55742±6348.1·S/μm2、平均光密度304.04±100.1、积分光密度18219±3548.9)比较均有显著统计学意义(均为P<0.05),模型组Nissl小体总面积、平均光密度、积分光密度与给药组(总面积46406±5989.3·S/μm2,平均光密度251.05±77.41,积分光密度16899±4040.6)比较,差异均有统计学意义(均P<0.05)。结论:补肾活血中药通过增强尼氏小体的表达、改善神经元细胞超微结构而促进EIOP大鼠初级视皮质损伤的修复。
Objective: To observe the effect of Bushen Huoxue Chinese medicine on the damage of primary visual cortex Nissl-body in rats with chronic intraocular pressure (EIOP) model, and to explore its mechanism. Methods: The model of chronic obstructive sleep apnea syndrome (EIOP) was established by the method of branching and closing the sclera vein and dissecting the three scleral veins of rats. The models were randomly divided into three groups: blank group, model group and administration group. The rats were sacrificed at the end of the 8th week, and the effects of intraocular pressure (IOP), Nissl body of primary visual cortex (PVC), ultrastructure of neurons influences. Results: The intraocular pressure (IOP) was significantly increased in this experiment (P <0.01), which was significantly different from that before model establishment (P <0.01). The pathological section of PVC Quantitative analysis showed that the total area of Nissl bodies in model group was 34941 ± 8482.1 · S / μm2, the average optical density was 152.8 ± 27.97, the integrated optical density was 11993 ± 3084.8, which was significantly higher than that of blank group (total area 55742 ± 6348.1 · S / μm2, Density of 304.04 ± 100.1, integral optical density of 18219 ± 3548.9) (all P <0.05). The total area, average optical density and integral optical density of Nissl body in the model group were significantly higher than those in the administration group (total area 46406 ± 5989.3 · S / μm2, average optical density of 251.05 ± 77.41, integral optical density of 16899 ± 4040.6), the difference was statistically significant (all P <0.05). Conclusion: Bushen Huoxue Herbs can promote the repair of primary visual cortex injury in EIOP rats by enhancing the expression of Nissl body and improving the ultrastructure of neurons.
