目的:制备与分析抗可提取性核抗原(ENA)单克隆抗体;方法:以ENA为抗原,免疫BALB/c小鼠,取其脾细胞与小鼠Sp2/0骨髓瘤细胞融合,经反复筛选和再克隆化,建立能稳定分泌抗ENA单克隆抗体(McAbs)的杂交瘤细胞株;结果:共获得11株能稳定分泌抗ENA的杂交瘤细胞株。对其中的5种单克隆抗体(IG5,2E5,2C6,4C10和4E6)进行了初步分析,结果如下:小鼠腹水抗体效价范围2×10-5~3×10-6;免疫球蛋白亚类鉴定,1株为IgG2a,1株为IgG2b,其余3株均为IgG1;结论:ENA作为弱免疫原可用于制备单克隆抗体
OBJECTIVE: To prepare and analyze anti-extractable monoclonal antibody (ENA) monoclonal antibody.Methods: BALB / c mice were immunized with ENA as an antigen and their splenocytes were fused with mouse Sp2 / 0 myeloma cells. After repeated screening And then cloned to establish a hybridoma cell line that can stably secrete anti-ENA monoclonal antibodies (McAbs). Results: A total of 11 hybridoma cell lines that can stably secrete anti-ENA were obtained. The five monoclonal antibodies (IG5, 2E5, 2C6, 4C10 and 4E6) were analyzed preliminarily. The results were as follows: the mouse ascites antibody titer ranged from 2 × 10-5 to 3 × 10-6; the immunoglobulin subunit One strain was IgG2a, the other was IgG2b. The other three strains were all IgG1. Conclusion: ENA can be used as a weak immunogen to prepare monoclonal antibodies
