广藿香挥发油对青蒿酯钠抗伯氏疟原虫的增效作用和对抗青蒿酯钠伯氏疟原虫的逆转抗性作用

[目的 ]研究广藿香挥发油 (B)与青蒿酯钠 (SA)联合用药对伯氏疟原虫 (P.b)正常株 (N)及抗青蒿酯钠株 (R)的抗疟增效作用和对 R的逆转抗性作用。[方法 ]采用 4天抑制试验法对分别感染 N和 R小鼠进行抗疟实验 ,以两药等效剂量配伍 ,用直线回归法计算其抑制原虫 5 0 %的药物剂量 (SD5 0 )。[结果 ]B单独用药 ,N:SD5 0 =87.6 4± 19.5 8(GKD) ,R:SD5 0 =43.2 4± 7.71(GKD) ;SA单独用药 ,N:SD5 0 =0 .88± 0 .0 1(MGKD) ,R:SD5 0 =2 7.6 9± 0 .93(MGKD)。B与 SA联合用药 ,N:B的 SD5 0 =36 .89± 4.5 7(GKD) ,SA的 SD5 0 =0 .39± 0 .0 5 (MGKD) ;R的 SD5 0 =7.40± 1.30 (GKD) ,SA的 SD5 0 =4.2 1± 0 .74(MGKD)。联合用药 B对 SA的增效指数 ,N、R分别为 2 .2和 6 .6 ,B对 R的逆转抗性倍数为 6 .6。相对逆转率为 87.6 %。[结论 ]采用 B和 SA联合用药对 P.b的抑制作用具有增效作用 ,能逆转 R对 SA的抗性及延缓 N对 SA抗性的发生。 [Objective] The research aimed to study the anti-malarial synergism effect of combined application of volatile oil of Pogostemon cablin (B) and artesunate sodium (SA) on normal strain of Plasmodium berghei (P) and anti-artemisinin sodium (R) And reversal of R resistance. [Method] The 4-day inhibition test was used to test malaria resistance in mice infected with N and R respectively. The drug dose (50% inhibition) of protozoal was calculated by linear regression with the equivalent dose of two drugs. [Results] B alone, N: SD5 0 = 87.6 4 ± 19.5 8 (GKD), R: SD5 0 = 43.2 4 ± 7.71 (GKD); SA alone, N: SD5 0 = 0.88 ± 0 .0 1 (MGKD), R: SD5 0 = 2 7.69 ± 0.93 (MGKD). B with SA, SD5 of N: B = 36.89 ± 4.57 (GKD), SA of SD5 0 = 0.39 ± 0.05 (MGKD); R of SD5 0 = 7.40 ± 1.30 (GKD ), SD5 0 of SA = 4.2 1 ± 0 .74 (MGKD). The synergistic index of the combination of B with SA, N, R were 2.2 and 6.6, respectively, and the ratio of B to R was -6.6. The relative reversal rate was 87.6%. [Conclusion] The combination of B and SA could inhibit the inhibition of P.b, which could reverse the resistance of R to SA and delay the occurrence of N against SA.
