目的系统分析ndrg2在胎前脑组织中表达分布的时空特点,以期明确ndrg2在胎前脑脑功能区的组织分布、细胞定位及表达趋势,为进一步研究其功能提供实验依据。方法收集引产4 ̄7月胎脑组织标本,分别进行组织石蜡切片和总RNA提取,应用免疫组化方法和RT-PCR技术检测ndrg2在蛋白质及mRNA表达水平变化规律。结果免疫组化结果表明,在上述胎前脑脑区神经元细胞胞浆中均有ndrg2蛋白不同程度的表达。RT-PCR结果显示,ndrg2mRNA在正常胎脑各脑区均有表达;随脑组织发育成熟,ndrg2表达水平增高,4月胎脑中海马ndrg2表达水平较高,而到7月额叶、顶叶则表达增强。以上结果表明,ndrg2基因在不同脑功能区广泛表达,但不同时期、不同胎脑功能区域表达有明显区别。结论在脑发育的过程中,ndrg2表达呈一定时空特点,ndrg2可能对神经细胞发育成熟有一定影响。
Objective To analyze the spatial and temporal characteristics of ndrg2 expression and distribution in fetal forebrain tissues so as to clarify the tissue distribution, cell location and expression trend of ndrg2 in fetal brain forebrain brain regions, and provide experimental evidence for further study of its function. Methods Fetal brain tissue samples were collected from April to July. Tissue paraffin sections and total RNA were extracted respectively. The expression of ndrg2 protein and mRNA was detected by immunohistochemistry and RT-PCR. Results Immunohistochemical results showed that there were different levels of ndrg2 protein expression in the cytoplasm of neurons in the fetal brain regions. The results of RT-PCR showed that ndrg2 mRNA was expressed in all brain regions of normal fetal brain. The expression of ndrg2 increased with the development of brain tissue. The level of ndrg2 in hippocampus was higher in April, The expression is enhanced. The above results show that ndrg2 gene is widely expressed in different brain function areas, but at different times, different fetal brain function areas were significantly different. Conclusion During brain development, ndrg2 expression has certain spatial and temporal characteristics, and ndrg2 may have some influence on the development of neural cells.
