目的:建立芫花药材中芫花素与芫花酯甲含量的测定方法。方法:采用高效液相色谱切换波长法同时测定芫花素与芫花酯甲的含量。色谱条件为Diamonsil C18色谱柱(250mm×4.6mm,5μm);流动相:甲醇-水(84:16);检测波长:前14min为338nm,14min后为232nm;柱温:25℃;流速:1mL/min。结果:此方法线性良好,芫花素与芫花酯甲的平均加样回收率分别为97.7%,97.0%,RSD分别为2.28%,1.64%(n=5)。结论:该方法简便、可靠、重复性好,可用于芫花药材中芫花素与芫花酯甲的同时检测。
Objective: To establish a method for the determination of genkwanin and yuanhuacine in Chinese traditional medicine. Methods: Simultaneous determination of genkwanin and yuanhuacine by high performance liquid chromatography with switched wavelength method. The chromatographic conditions were Diamonsil C18 column (250 mm × 4.6 mm, 5 μm); mobile phase: methanol-water (84:16); detection wavelength: 338 nm in the first 14 min and 232 nm in 14 min; / min. Results: The method was linear and the average recoveries of genkwanin and yuanhuacine were 97.7% and 97.0%, respectively. The RSDs were 2.28% and 1.64%, respectively (n = 5). Conclusion: The method is simple, reliable and reproducible. It can be used for simultaneous detection of genkwanin and yuanhuacine in Daphne genkwa.
