目的 :通过苦参碱对大鼠贮脂细胞株HSC -T6增殖及胶原、透明质酸合成的影响作用 ,研究苦参碱抗肝纤维化的作用机理。方法 :采用大鼠肝贮脂细胞株 ,以细胞增殖抑制率为指标 ,用噻唑蓝 (MTT)法 ,测定药物对大鼠贮脂细胞株HSC -T6增殖的影响 ;3H -脯氨酸掺入法 ,测定药物对HSC -T6胶原合成的影响 ;放免法测定药物对HSC -T6合成透明质酸的作用。结果 :苦参碱对大鼠贮脂细胞株HSC -T6增殖 ,胶原和透明质酸合成的抑制率随药物浓度增加而升高 ,呈药物浓度依赖关系。结论 :苦参碱通过抑制贮脂细胞增殖及细胞外基质的合成 ,发挥抗肝纤维化的作用
Objective :To study the effect of matrine on liver fibrosis by studying the effect of matrine on the proliferation of rat fat-storing cell line HSC-T6 and the synthesis of collagen and hyaluronic acid. METHODS: Rat hepatocyte-secreting cell line was used to observe the proliferation inhibition rate of cells. The effect of drug on the proliferation of rat fat-storing cell line HSC-T6 was determined by MTT assay; 3H-proline spiked Methods: The effect of drugs on the synthesis of HSC-T6 collagen was determined; radioimmunoassay was used to determine the effect of drugs on HSC-T6 synthetic hyaluronic acid. RESULTS : Matrine inhibited the proliferation of HSC-T6 cells and inhibited the synthesis of collagen and hyaluronic acid. The inhibitory rate of matrine increased with the increase of drug concentration, showing a drug concentration dependence. Conclusion: Matrine exerts anti-fibrosis effect by inhibiting the proliferation of extracellular lipids and the synthesis of extracellular matrix.
